Could I get the same effect as that described by replacing the microphone with someone totally deaf in one ear?

If I play an old mono song on my stereo and reverse the wires (phase) of one speaker, I can find a spot where a mono microphone will fail to pick up any sound (as long as the speakers are angled correctly and the room isn't too reflective).

Yes that would work. I think the phenomenon is known as destructive interference, where two waves interact to cancel each other out. This would occur regardless of the type of detector that you are using.

Do read and consider the "directionality" of microphones, ears and the fact that (if said half deaf person were "not on top of a flag pole" but in a room) a great deal of what is heard is reflected sound thus following a different path length which negates  much of the destructive interference.

The null is not as sharp as one might expect because much of the sound reaching our ear drums is conducted through the bone of the skull. Now, if you could find someone deaf in one ear and with a very small head................
